Polymerase Chain Reaction, Restriction Fragment Length Polymorphism and Plasmid Profile Analysis of Salmonella isolates of Chicken Egg Origin

ABSTRACT
A total of 27 Salmonella isolates belonging to 4 different serotypes isolated from chicken eggs were subjected to polymerase chain reaction (PCR), PCR-RFLP (restriction fragment length polymorphism) and plasmid profiling. All the isolates were positive for hisJ gene and amplified a specific fragment of 496 bp. RE digestion of PCR product using RsaI, HindIII and HaeIII revealed similar RFLP pattern indicating no polymorphism among the isolates. All the Salmonella isolates harbored more than one plasmid with seven different plasmid profiles including 2 to 4 plasmids with approximate molecular size ranging from 3.2 kb to 23 kb.Keywords: Polymerase chain reaction, plasmid, RFLP, Salmonella
