Purification of Salmonella Enterotoxin and its Detection in Field Salmonella Strains by Dot-ELISA

ABSTRACT

Salmonella enterotoxin (Stn) was purified from the cell free culture supernatant of Salmonella Typhimurium by salt precipitation, dialysis and successive gel filtration chromatography on Sephdex G-100 and G-200. The final elution profile following chromatography revealed two peaks and the enterotoxic activity was detected in the first peak. SDS-PAGE of the purified toxin showed a single band of protein with a molecular weight of 60 kDa. Antiserum was raised in the rabbits against this purified toxin and was used for detection of Stn in field Salmonella strains by dot-ELISA. The Stn could be detected in 88 (92.6%) of the 95 isolates. The test was found to be sensitive, simple and rapid and can be used to detect Stn for characterization of Salmonella in the public health laboratories.
