ABSTRACT
Indirect haemagglutination (IHA) with various antigens viz., CFT antigen, mallein PPD and recombinant antigen was standardized for the diagnosis of glanders. The optimum dilution of above antigens was found as 1/10, 1/5 and 1/18 containing 10.96 μg/ml, 23.30 μg/ml and 3.21 μg/ml protein, respectively. The tanned formalinised sheep RBCs were found better when used on the same day of coating antigens. The albumin bovine serum-PBS has been found as better diluent than normal rabbit serum-PBS. An IHA titre of 320 and above was considered as significant titre for serodiagnosis of glanders in the present study. Analysis of 248 field equine sera proved that IHA with CFT antigen was better than IHA with mallein PPD and recombinant antigen, as it had a higher sensitivity (63.64%), specificity (99.16%), positive predictive value (77.78%), negative predictive value (98.33%) with 97.58% accuracy and 2.4% error of prediction.Keywords: CFT antigen, glanders, indirect haemagglutination, mallein PPD, recombinant antigen
